What volume should I use for DNA quantitation when reading at 260 nm?
Using the Corning 3635 and similar UV transmissable microplates, the minimum volume is 100 µL. Using the Corning 3679 half area microplates, the minimum volume is 50 µL. Using 384-well UV microplates, the minimum volume is 25 µL. For the most accurate readings, we recommend 200, 100, and 50 µL or more respectively.
