How do I optimize the instrument for reading assays with live cells (cellular assays) on the bottom of the wells?
- Use the Optics position bottom setting.
- If possible, use plates whose bottom plastic has been optimized for bottom reading using fluorescence and match with the correct plate in the database.
- Scale to high well helps to rapidly find the optimum PMT voltage for your assay.
- Choose the appropriate filters and make sure the bandpasses are not too close or the software will provide a warning.
- Read using the default settings under Options although 100 reads could provide better CVs.
- If exciting-detecting at 485/20-528/20, remove phenol red from any media.
- Cover the plate if sterility needs to be maintained.
- BioTek's large optical bottom probe and high quality filters provide excellent signal to noise data for cellular assays.
